Abstract

The beta -lactoglobulin genotype is known to be associated with the content of casein and ratio of casein to total protein (casein number) in milk (AA < AB < BB ), possibly because of mutations in regulatory elements in the 5'-flanking region of the beta -lactoglobulin gene. In an effort to explain the difference in expression between the A and B genetic variants, nucleotide variation was studied at positions -435 (R10) and -462 (R11) relative to the transcription start point in the 5'-flanking region of the bovine beta -lactoglobulin gene. A total of 361 cows from three different breeds in Sweden were genotyped. Complete linkage disequilibrium was found between the polymorphisms at positions R10 and R11 and the polymorphism at codon 64 that was utilized for genotyping of the beta -lactoglobulin A and B genetic variants. Due to the complete linkage disequilibrium, the polymorphisms at positions R10 and R11 did not provide any additional information as regards the difference in casein number between the A and B genetic variants.
