Separation and identification of cytokinins using combined capillary liquid chromatography/mass spectrometry

Abstract

Non-derivatized cytokinins were analysed by liquid chromatography/mass spectrometry (LC/MS). The effluent from a capillary reversed-phase high-performance liquid chromatography column was introduced into the ion source of a double-focusing mass spectrometer via a frit interface, and ions were generated by fast atom bombardment (FAB) with 1% glycerol in the mobile phase acting as a matrix. Positive FAB spectra were obtained for base, riboside, ribotide and glucoside forms of cytokinins. The spectra were found to provide useful information for identification and structural elucidation of cytokinins. The LC/MS system was used to identify iso-pentenyladenosine in a purified extract from Norway spruce (Picea abies) needles. Quantitative analysis of iso-pentenyladenosine using (H-2(6))isopentenyladenosine as internal standard indicated levels of 1.2 ng g-1 fresh weight.
