Sammanfattning

The rapid freeze and freeze substitution method for fixing the differentiating xylem has given us new information on cell wall assembly. Hemicelluloses are synthesized in the Golgi apparatus and transported to the inner surface of the developing cell wall by exocytosis of Golgi vesicles. As a result of hemicelluloses deposition, the inner surface of the developing cell walls is always coated with a thin layer composed of hemicelluloses and a significant amount of water. Lignification excludes water from nonlignified and lignifying cell walls. Immunocytochemistry reveals not only the localization of enzymes involved in the biosynthesis of cell wall components, but also the localization of hemicelluloses and lignins within the cell wall. Biochemical investigations suggest that the mechanism of monolignols transportation from the cell to the developing cell walls is by passing through the plasma membrane.
